Salmonella Testing Methods

Rapid Salmonella PCR
Polymerase Chain Reaction (PCR) technology amplifies specific DNA fragments unique to the Salmonella genus. The process begins with an enrichment phase to revive injured cells in the food matrix, followed by lysis to release DNA. The thermal cycler then targets and replicates the invA gene or other specific genetic markers.

Advantages:This method offers high sensitivity and specificity with a rapid turnaround time. Negative results are typically available within 24 to 48 hours, allowing for the faster release of perishable food products.
Traditional Salmonella Culture
This is the gold standard for microbiological analysis. It involves a multi-step process: pre-enrichment in non-selective broth to recover stressed bacteria, selective enrichment to inhibit background flora, and isolation on differential plating media like XLD, HE, or Bismuth Sulfite agar.
FDA BAM Chapter 5 | ISO 6579-1:2017.
Advantages:While longer (4-5 days), cultural methods allow for the isolation of the living organism. This is essential for regulatory disputes and outbreak investigations.
Biochemical Confirmation
Any sample that tests presumptive positive via PCR or screening methods must undergo confirmation. This involves analyzing the metabolic profile of the isolate (sugar fermentation, hydrogen sulfide production) and checking for agglutination with specific Salmonella O and H antisera.
